Abstract
The only effective method of identification of beekeeping products by botanical and geographical origin is to study the specificity of pollen grains in the composition of the said products. Identification of pollen grains by morphological characteristics and their percentage in the product allows to establish a botanical honey variety (black locust, linden). Light and electron microscopy techniques are used to study the peculiarities of the morphological structure of pollen grains. The lack of detailed identification of pollen grains of many species of entomophilous plants of Ukraine makes the study of their morphology relevant. The purpose of the work was to test and adapt known electronic scanning microscopy methods to identify pollen grains on equipment available in Ukraine. The material was herbarium specimens of different species of entomophilic plants collected under the conditions of the Kiev region. Plant identification and sample preparation were performed at the laboratories of the Department of Standardization and Certification of Agricultural Products. Tests of the methods were performed at the laboratory of electron microscopy LLC "NanoMedTech" using an electron scanning microscope Tescan Mira 3 LMU, precision etching system and coating Gatan Pecs 682. It was found that the use of electron microscopy techniques adapted to the existing equipment for the preparation of samples based on 2,2-dimethoxypropane will allow to study the morphology of pollen grains of entomophilous plants of Ukraine, this will further enable them to be identified in beekeeping products. Images obtained by the method of preparation of the sample using fixation based on glutaraldehyde, showed compliance with the shape but deformed surface of the grain. The shape and structure of the pollen grain surface was preserved in the preparation of the sample by the method using 2,2-dimethoxypropane. The use of an adapted sampling technique based on pre-treatment with 2,2-dimethoxypropane, dehydration and drying at a critical point of CO2, followed by scanning electron microscopy methods enables the identification of pollen grains of honey plants. In the future, it is planned to supplement the images obtained with the international PalDat online database, which will allow to establish botanical and geographical origin of bee products from Ukraine, which is sold abroad.
